During the Second World War the airfield was used by the United States Army Air Forces Sixth Air Force from 1942 until 1944, operating as Camaguey Air Base. The 25th Bombardment Group's 417th Bombardment Squadron flew B-18 Bolo bombers from the field between 13 April 1942 and August 1943, carrying out antisubmarine patrols over the northern Caribbean, while the base also supported air-sea rescue missions flown by the 1st Rescue Squadron. From 1 January 1943 the USAAF ran postal operations for Camaguey through the Army Post Office in Miami, and the United States Navy separately used the Fleet Post Office, Atlantic, in New York for its own mail handling at the base. In the post-war era the airfield served as a base for the Cuban Revolutionary Armed Forces, hosting the 3685th Regiment and two general-purpose transport squadrons operating Mil Mi-17 helicopters. The airport was closed to traffic from March to October 2020 because of the COVID-19 pandemic.
The airport continues to operate as an international gateway for Camagüey and the Santa Lucía resort area, with scheduled and charter flights including American Airlines services to Miami and seasonal Viva flights to Cancún. It remains under ECASA management and handles both domestic and international traffic.
